أعلن باحثون في جامعة واشنطن بالولايات المتحدة، وبالتعاون مع جامعة نورثويسترن، عن تطوير علاج غير تدخلّي باستخدام قطرات أنف نانوية، قادرة على توصيل جزيئات محفزة للمناعة مباشرة إلى الدماغ لمحاربة الورم الأرومي الدبقي، أحد أخطر أنواع سرطان الدماغ.

في التجارب التي نُشرت في دورية علمية رائدة، استخدم الفريق ما يُعرف بـSpherical Nucleic Acids (جسيمات نووية كروية)، جزيئات نانوية ذات نواة ذهبية مغطاة بحمض نووي يُبرمج لتنشيط مسار مناعي داخل الدماغ يُعرف بـSTING pathway، وهو المسار نفسه الذي يُشغّل الاستجابة المناعية عند اكتشاف خلايا غريبة.

الميزة الأساسية، بحسب الباحثين، أن هذه القطرات تدخل من خلال الأنف، فتسلك المسار العصبي المباشر نحو الدماغ، متجاوزة الحاجز الدموي الدماغي الذي غالباً يمنع الأدوية من الوصول إلى الأنسجة الدماغية.

عند وصول الجسيمات إلى الدماغ، حفّزت استجابة مناعية قوية ضد الورم؛ وخلال تجارب على فئران مصابة بالورم الأرومي الدبقي، أشارت النتائج إلى أنه في بعض الحالات، بعد جرعة أو اثنتين من القطرات، تمت إزالة الورم تماماً مع تنشيط خلايا المناعة محلياً دون انتشار التأثير ضاراً في باقي الجسم.

هذا المنحى العلاجِي غير التداخلي يُعَدّ ثورة محتملة في علاج أورام الدماغ، لأن السبل التقليدية، الجراحة، الإشعاع، الكيميائي، غالباً ما تفشل في استئصال الورم كاملاً، وتتعرض لأنسجة دماغ سليمة، أو لا تستطيع اختراق الحاجز الدموي الدماغي بفعالية كافية.

الباحثون يؤكدون أن هذه الطريقة — ببساطتها (مثل بخاخ أنف) — تمثل تحوّلاً جذرياً، خصوصاً مع قدرة القطرة على تحويل ما يُعرف بورم «بارد» (لا يثير المناعة) إلى ورم «ساخن» (يُستهدف من الجهاز المناعي).

مع ذلك، حتى الآن النتائج إيجابية في تجارب ما قبل السريرية (الفئران). لا يزال أمام العلماء وقت طويل قبل أن يُتاح العلاج للبشر، لأن هناك حاجة لتقييم الأمان طويل الأمد، إمكانية التوسع الإنتاجي، وفعالية مع مختلف أنواع الورم ومراحله.

Researchers at the University of Washington in the United States, in collaboration with Northwestern University, have announced the development of a non-invasive treatment using nanoscale nasal drops, capable of delivering immune-stimulating particles directly to the brain to combat glioblastoma, one of the most dangerous types of brain cancer.

In experiments published in a leading scientific journal, the team used what is known as Spherical Nucleic Acids, nanoscale particles with a gold core coated with nucleic acid programmed to activate an immune pathway in the brain known as the STING pathway, which is the same pathway that triggers the immune response upon detecting foreign cells.

The key advantage, according to the researchers, is that these drops enter through the nose, taking a direct neural route to the brain, bypassing the blood-brain barrier that often prevents drugs from reaching brain tissues.

Upon reaching the brain, the particles triggered a strong immune response against the tumor; and during experiments on mice with glioblastoma, results indicated that in some cases, after one or two doses of the drops, the tumor was completely removed with local activation of immune cells without harmful effects spreading to the rest of the body.

This non-invasive therapeutic approach is considered a potential revolution in the treatment of brain tumors, as traditional methods—surgery, radiation, chemotherapy—often fail to completely remove the tumor, affect healthy brain tissues, or cannot effectively penetrate the blood-brain barrier.

The researchers emphasize that this method—due to its simplicity (like a nasal spray)—represents a radical shift, especially with the drop's ability to transform what is known as a "cold" tumor (which does not provoke an immune response) into a "hot" tumor (targeted by the immune system).

However, so far the results are positive in preclinical trials (in mice). Scientists still have a long way to go before the treatment is available for humans, as there is a need to assess long-term safety, production scalability, and effectiveness against various types and stages of tumors.
